Genetically Engineered Mice

The Genetically Engineered Models (GEM) Collection contains nearly 400 hundred proprietary GEM lines with all of them accessible to Nonprofit organizations and nearly half of them available to For Profit organizations. Models are available after an MTA has been signed between the originator and host institutions (see terms of use). The GEM collection is one of several Taconic repositories of rodent models relevant to life science research. It represents a wide range of protein target classes and includes GEM of relevance to several disease areas, such as cardiovascular, diabetes/ endocrinology, neuroscience, oncology, respiratory, and immunology. Scientists seeking GEM lines can use the search button on the upper right on this page to search all of Taconic's collections (e.g. GEM Collection, Taconic Transgenic Models, Taconic Knockout Repository), providing them access to one of the world's most extensive repositories of genetically engineered mice and rat models.

The GEM Collection contains mainly conditional (cKO) and constitutive (KO) Knock Out mice, transgenic (Tg) over-expressing lines, conditional (cTTG) and constitutive targeted transgenesis (TTG) lines. The vast majority of the lines are in a C57BL/6 genetic background.

Investigators are welcome to search the Taconic website for gene(s) of interest. The webpage for each model will provide the allele diagram, detailed descriptions of how the GEM line has been generated, as well as terms of use, deliverables, and pricing. Investigators from research institutions can learn how to access models from the GEM Collection and may find information about deliverables and pricing. Investigators may also inquire with Taconic's customer service with any questions.
Nonprofit users may purchase GEM Collection models by signing the appropriate agreement posted on the model page and providing a PO. Depending on the model, the agreement is either the GEM Collection Academic Terms of Sale or a Materials Transfer Agreement.

For models distributed under a Materials Transfer Agreement (only), the key MTA terms are:

  • Rodent Model Materials are provided only for Research performed by Requestor, not on behalf of a Third Party, and may not be transferred to any Third Party without Merck Sharp & Dohme Corp.'s (MSD) prior written consent.
  • A work plan describing the Requestors intended Research scope is required, and, following execution of the Agreement, such work plan may only be changed or expanded through written notice to MSD.
  • MSD retains ownership of Rodent Model Materials, including components of such contained in any Modified Derivatives.
  • Modified Derivatives may be made and used only by the Requestor in accordance with the Work Plan and terms of the MTA. Modified Derivatives may be transferred to Third Parties only upon amendment of the MTA and establishment of an MTA between such Third Party and MSD.
  • The Rodent Model Materials and Modified Derivatives and any information provided may only be used for the Research and may not be used to develop a commercial product. They may only be transferred upon amendment of the MTA and establishment of an MTA between any receiving Third Party and MSD.
  • Written report of Research results must be provided to MSD, under terms of confidentiality for MSD's internal use only.
  • Inventions and discoveries resulting from the Research must be reported to MSD. MSD may elect to establish a non-exclusive or exclusive license, to be negotiated on reasonable terms.
  • MSD has a worldwide, royalty-free, non-exclusive, sublicensable right to use Modified Derivatives.
  • No further use of the Rodent Model Materials and Modified Derivatives may be made after the expiration or termination of the MTA.
  • For some Rodent Models a Third-Party License may be required.
Academic customers must provide a scientific workplan within the MTA — answering 4 basic questions:

  1. For what research will the requested mice be used?
    Example: To Study the role Histone Deacetylases in cardiac growth under both physiological and pathophysiological conditions, heart failure in particular

  2. How will the mice be used experimentally?

    • Challenge the hearts of
      • mice with a global deletion of the HDAC xx gene (KO)
      • mice that lack HDAC xx gene specifically in cardiomyocytes (cKO)
    • Cardiac function will be assessed by left ventricular pressure-volume conductance catheterization and transthoracic echocardiography.

  3. What data is expected to be generated and in what timeframe (1 year, 18 months, 2 years, etc.)
    Example: Phenotypic data on both Hdac xx KO and cKO in 2 year time frame.

  4. What translational aspects do you envision your data may inform or lead to (better understanding of mechanism, candidate drug targets, better animal model of human disease etc.)?
    Example: To dissect the roles of different HDACs for pathological cardiac remodeling and thus the development of heart failure.
A workplan is not needed for models distributed under the Academic Terms of Sale.
Commercial users may purchase available GEM Collection models by signing the GEM Collection Commercial Terms of Sale and providing a PO. The GEM Collection Commercial Terms of Sale grant perpetual use rights.
Q. What health standard is guaranteed for the mice delivered?
A. Murine Pathogen Free™ (MPF™). Mice at a more restrictive health standard may be delivered upon request and at additional cost.

Q. How many mice will the customer receive?
A. The customer will receive at least 4 heterozygous mice. In rare cases that the cryopreserved material also contains a recombinase gene, this may be reduced to 1-2 heterozygous mouse.

Q. Can Taconic send cryopreserved materials such as sperm or embryos to the customer in lieu of live mice?
A. No. Cryopreserved materials are the property of Taconic or the Sponsor and may not be provided directly to customers.

Q. What happens if none of the mice sent are fertile?
A. Taconic cannot warranty individual mice for fertility. Mice are provided within the optimal breeding age range. Heterozygous mutants are less likely to have fertility problems compared to homozygous mice. Taconic recommends that customers consider doing at least one round of expansion by breeding the received heterozygous mice to wild type prior to intracrossing to homozygosity to reduce the chance of problems propagating the mice received. Taconic will discuss environmental and husbandry factors with the customer to determine whether there may be an external problem.

Q. What happens if the mice die in shipment?
A. Customers must inform Taconic in the event the mice die during transit or within 24 hours of receipt. Taconic will attempt to replace the shipment using mice available. Customers should unpack and inspect all shipments of mice immediately after arrival. Mice that arrive dead or in poor health should be reported within 3 days by the receiving institution. Taconic does not have control over housing and environmental conditions in recipient facilities and thus cannot be held responsible for mice that die more than 24 hours after receipt.

Q. If a customer wishes to perform a rapid expansion package to expand a line, do they still need to separately pay the cryorecovery package fee?
A. The price shown on the model page is due upon completion of cryorecovery. Any additional services such as a Rapid expansion are an additional service which will be billed in addition to the basic package.

Q. Can GEM Collection lines be crossbred or further modified?
A. The GEM Collection Commercial Terms of Sale and Academic Terms of Sale permit breeding, crossbreeding and other further modifications of the model. The academic MTA permits breeding, but crossbreeding or further modifications may only be done to perform the work described in the scientific work plan. Refer to the specific model page to determine whether it is available under the Academic Terms of Sale or academic MTA.

Q. What is the timeline to receive deliverables?
A. Once the appropriate agreement has been executed and the customer has provided a PO, the timeline is approximately 14-16 weeks until live mice are ready for shipment. Mice are provided at 6-12 weeks, which is the optimal breeding age.

Q. How long is the term of the agreement?
A. The GEM Collection Commercial Terms of Sale grant perpetual use rights to commercial customers. The Academic GEM Collection Terms of Sale grant perpetual use rights to nonprofit customers. The academic MTA has a three-year term. Renewal of the academic MTA is at the discretion of Merck Sharp & Dohme ("MSD"). The fee for such renewals is set by MSD.
